Role of prolactin on Leydig, Sertoli and germ cellular neutral lipids in bonnet monkeys, Macaca radiata.

چکیده
To elucidate the specific influence of prolactin on neutral lipids in Leydig, Sertoli and germ cell compartments of the testis in immature and mature monkeys, the present study was carried out by injecting ovine prolactin (oPRL) (1 mg/kg body weight/twice daily for 10 days ip), to both age groups. Similarly, bromocryptine (an ergot alkaloid which inhibits prolactin secretion) was given to other sets of immature and mature monkeys (1 mg/kg body weight/twice daily for 10 days ip) to induce hypoprolactinemia. It was observed that after oPRL administration the total lipid accumulated in the germ cells of immature and mature monkeys. Total lipid was markedly decreased in the Leydig cells of mature monkeys only. But no such influence of PRL was evident in the Leydig cells of immature monkeys, suggesting an age-dependent effect of PRL on the Leydig cells. The increase in total lipid in the germ cells following PRL treatment was contributed by mono, di- and triacyl glycerols and free cholesterol. However, an opposite effect of PRL was evident in the Leydig cells of mature monkeys, where the cholesterols and glyceride fractions registered a decrease. The reduced cholesterol fractions in the Leydig cells following PRL treatment suggests the utilization of cholesterol for steroidogenesis. Sertoli cells were found to be comparatively resistant to change in PRL status. Bromocryptine treatment brought about the opposite effect of PRL in almost all parameters studied in both immature and mature monkeys. In general, these findings with prolactin suggests that PRL has a specific and definite influence on testicular neutral lipids and the response of different cellular compartments was found to vary.

Effect of Passive Immunization with Specific Anti-FSH on Tes- ticular Lipids and Some Lipogenic Enzymes of Adult Bonnet Monkeys
The effect of anti-follicular stimulating hormone (FSH) on testicular lipid and the specific activity of testicular enzymes of the isocitrate dehydrogenase (ICDH), pyruvate/malate dehydrogenase (MDH) and malic enzyme involved in lipogenesis were studied in mature bonnet monkeys, Macaca radiata. Immunization of monkey with anti-FSH for 24 days did not produce any significant changes in the body ...
متن کاملIsolation and characterization of Leydig cells from adult bonnet monkeys (Macaca radiata): evidence for low steroidogenic capacity in monkey Leydig cells in contrast to rat Leydig cells.
Most of the available information on Leydig cells has been obtained using a rodent model system. With an objective to extend the observations made with rat Leydig cells (RLCs) to primates, a method has been developed to isolate Leydig cells from monkey (Macaca radiata) testis. Enzymatic dissociation of monkey testis followed by Percoll-gradient fractionation of the interstitial cells resulted i...
متن کاملP-7: Testosterone and Gonadotrophin Hormones Plasma Concentration-Induced Changes in Sertoli and Leydig Cells Number in Hyperprolactinemic Mature Male Mice
Background: Hyperprolactinemia is a particular concern in the treatment of schizophrenic patients. The side effects associated with high prolactin levels can have a negative impact on GnRH pulsatile secretion and, consequently LH, FSH and testosterone pulsatility and follows by, on special testicular cells that have these hormon’s receptors such as sertoli and leydig cells. Quantification the d...
متن کاملRegulation of testosterone rhythmicity by gonadotropins in bonnet monkeys (Macaca radiata).
In an attempt to study the factor(s) that regulates production of nychthemeral testosterone surges in adult male bonnet monkeys (Macaca radiata), serum levels of testosterone, LH, FSH, and prolactin were monitored during a 24 h period. Only prolactin showed a significant increment in its levels coincident with that of the testosterone surge. The relationship between LH and testosterone producti...
متن کاملMorphological events in the primate endometrium in the presence of a preimplantation embryo, detected by the serum preimplantation factor bioassay.
BACKGROUND Hormonal modulation of the endometrium towards receptivity is well established; however, the role of embryonic stimuli in modulation of the endometrium prior to implantation, especially in primates, is unknown. The aim of the present study was to evaluate the endometrial histology when the embryo was present in its vicinity prior to implantation. METHODS Preimplantation factor (PIF...
